go through意为“穿过”，其中 through为介词，表示“从……的一边到另一边”，着重指从物体内部穿过或透过。
1) It took us a whole week to go through one of the great forests.
我们花了整整一星期才穿过一大森林。
2) The rain has gone through my coat.
雨水湿透了我的上衣。
补充: across 作介词时，也有“从……的一边到另一边”之意，但它着重指从物体表面横过。
我下了车，穿过一个门……

“in + 时间段” 意为“在……之后”，常与将来时连用，对其提问时用how soon。注意：in不能与时间点连用。
“after + 时间段”也可表示“在……之后”，但常与过去时连用；

What’s their difference in meaning
after还可与时间点连用，此时可用于将来时中。
